The development of super-resolved optical fluorescence microscopy methods has significantly advanced single-molecule tracking and imaging. The exceptional localization precision of just a few nanometers opens up new biophysical insights into the molecular mechanisms involved in neuronal and oncological diseases. Unlike ensemble-averaging measurements, single-molecule localization tracking and imaging at the nanoscale provide quantitative information on the kinetics of individual molecules within living cells and their respective interactions. Our modeling and understanding of these processes will be linked to a key case study focusing on chromatin-DNA organization and function.
